从人淋巴结中分离与纯化HMGN2蛋白,探讨HMGN2分子抗菌作用及其在淋巴结中的亚细胞分布。我们采用高效液相色谱分离纯化,琼脂糖弥散法筛选纯化组分的抗菌活性,对活性组分进行质谱分析测定精确分子量,进行Western-blot检测同源性。结果成功从人淋巴结中分离纯化出的活性蛋白,对致病性大肠杆菌54080有抗菌活性,质谱精确分子质量测定、Western-blot分析证明此活性蛋白即为HMGN2分子,免疫组化实验结果显示在淋巴结组织细胞细胞浆和细胞核均有HMGN2蛋白分布。从人淋巴结组织成功分离纯化出纯度较高、具有生物活性的HMGN2蛋白,可能参与了体内的天然免疫防御作用。
Isolation and purification of HMGN2 protein from human lymph nodes to explore the antibacterial effect of HMGN2 molecules and their subcellular distribution in lymph nodes. We use high performance liquid chromatography separation and purification, agarose screening screening purification components of antibacterial activity, the active component mass spectrometry determination of accurate molecular weight, Western-blot detection of homology. Results The active protein isolated and purified from human lymph node successfully showed antibacterial activity against pathogenic Escherichia coli 54080. The accurate mass spectrometry was confirmed by Western-blot analysis. The result showed that the active protein was HMGN2. The results of immunohistochemistry showed that Lymph node tissue cell cytoplasm and nucleus HMGN2 protein distribution. The successful purification and purification of HMGN2 protein with high purity and biological activity from human lymph node tissue may be involved in the natural immune defense in vivo.
