目的:检测抑癌基因RASSF1A在胃癌细胞株及组织中的表达情况,探讨RASSF1A在胃癌发生发展中的作用及临床意义。方法:用RT-PCR及Western blot检测细胞株、胃癌及癌旁组织中RASSF1A基因及蛋白的表达水平。结果:RASSF1A在低分化BGC-823细胞中的表达量明显低于中分化SGC-7901细胞,在胃癌中的表达阳性率(43.48%)显著低于癌旁组织(85.51%),且低分化胃癌中的表达低于中高分化,Ⅲ、Ⅳ期胃癌中的表达低于Ⅰ、Ⅱ期(P<0.05)。结论:在原发性胃癌组织中,RASSF1A的表达明显缺失或低下,且与肿瘤的分化程度及分期相关。
Objective: To detect the expression of tumor suppressor gene RASSF1A in gastric cancer cell lines and tissues, and to explore the role and clinical significance of RASSF1A in the development of gastric cancer. Methods: RT-PCR and Western blot were used to detect the expression of RASSF1A gene and protein in cell lines, gastric cancer and adjacent tissues. Results: The positive expression rate of RASSF1A in poorly differentiated BGC-823 cells was significantly lower than that in moderately differentiated SGC-7901 cells (43.48%, P <0.05), and poorly differentiated gastric carcinoma The expression in stage Ⅲ and Ⅳ gastric cancer was lower than that in stage Ⅰ and Ⅱ (P <0.05). Conclusion: The expression of RASSF1A is significantly decreased or decreased in primary gastric cancer, which is related to the degree of tumor differentiation and staging.
