目的:建立HPLC同时测定长白山牛皮杜鹃中芦丁、槲皮素及山柰酚含量的方法,并比较不同提取方法对其含量的影响。方法:采用Agilent Eclipse XDB-C18色谱柱(4.6 mm×250 mm,5μm);乙腈-0.1%磷酸水溶液进行梯度洗脱;检测波长为360 nm,流速为0.8 m L·min-1,柱温为30℃。结果:芦丁、槲皮素、山柰酚的在一定范围内线性关系均良好;平均加样回收率分别为99.9%(RSD 1.2%),99.2%(RSD 2.8%),101.8%(RSD 2.3%)。结论:该方法简便、快速、准确可靠。3种不同提取方法中以超声提取法所得黄酮类含量均最高。可用于长白山牛皮杜鹃的质量控制。
OBJECTIVE: To establish a HPLC method for simultaneous determination of rutin, quercetin and kaempferol in Rhododendron grosvenorii from Changbai Mountain. The effects of different extraction methods on the contents of rutin, quercetin and kaempferol were compared. METHODS: The gradient was eluted with Agilent Eclipse XDB-C18 column (4.6 mm × 250 mm, 5 μm) and acetonitrile-0.1% phosphoric acid aqueous solution. The detection wavelength was 360 nm and the flow rate was 0.8 m L · min-1. 30 ° C. Results: The linear relationship between rutin, quercetin and kaempferol was good within a certain range. The average recoveries were 99.9% (RSD 1.2%), 99.2% (RSD 2.8%) and 101.8% (RSD 2.3 %). Conclusion: The method is simple, rapid, accurate and reliable. The flavonoids content obtained by ultrasonic extraction was the highest among the three different extraction methods. It can be used for quality control of rhododendron in Changbai Mountain.
