目的探讨丹参酮ⅡA对大鼠心肌纤维化的保护作用。方法 SD大鼠60只采用腹主动脉缩窄术制备压力负荷增加心肌纤维化模型,术后存活中的24只大鼠随机分为3组,假手术组、模型组、丹参酮ⅡA组(20mg/(kg.d),每组各8只。术后4周造模成功并开始给药,疗程为4周,8周后检测心肌质量指数、心肌组织病理学、心肌羟脯氨酸及心肌组织转化生长因子-β1蛋白水平。结果模型组大鼠心肌质量指数、心肌羟脯氨酸及心肌组织中转化生长因子-β1蛋白水平明显高于假手术组与丹参酮ⅡA组(P<0.01)。结论丹参酮ⅡA可抑制心肌肥厚,减轻心肌纤维化,可能与下调心肌组织转化生长因子-β1表达有关。
Objective To explore the protective effect of tanshinone ⅡA on myocardial fibrosis in rats. Methods Twenty-four SD rats were randomly divided into three groups: sham operation group, model group, tanshinone Ⅱ A group (20mg / (kg.d), each group of 8. After 4 weeks of successful modeling and start the medication for 4 weeks, 8 weeks after the detection of myocardial mass index, myocardial histopathology, myocardial hydroxyproline and myocardial tissue transformation Β1 protein level in model group.Results The levels of myocardial mass index, hydroxyproline in myocardium and the level of transforming growth factor-β1 in myocardial tissue in model group were significantly higher than those in sham group and tanshinone ⅡA group (P0.01) .Conclusion Tanshinone ⅡA can inhibit cardiac hypertrophy and reduce myocardial fibrosis, which may be related to the down-regulation of the expression of transforming growth factor-β1 in myocardium.
