1目的 探讨人乳头瘤病毒 16 ,18型 (HPV16 ,18)感染及 ras基因突变与喉鳞癌的关系。2方法 应用聚合酶链反应 (PCR)和限制性片段长度多态性 (RFL P)分析技术 ,检测 40例喉鳞状细胞癌组织中 ras基因 (N-ras,K- ras,H- ras)第 12密码子的突变情况及 HPV16 ,18的感染情况。3结果 40例标本中检出 HPV16阳性 14例(35 % ) ,HPV18阳性 4例 (10 % )。 9例标本 ras基因发生突变 ,其中 N- ras6例 ,K- ras2例 ,H- ras1例。ras基因在HPV16阳性和阴性标本中的突变率分别为 11.11% (2 / 18)和 31.82 % (7/ 2 2 ) ,二者差异无显著意义 (χ2 =1.3917,P>0 .0 5 )。 4结论 HPV感染和 ras基因突变在喉鳞癌的发生发展过程中起重要作用。
1 Objective To investigate the relationship between human papillomavirus type 16 and 18 (HPV16, 18) infection and ras gene mutation and laryngeal squamous cell carcinoma. 2 Methods Polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) were used to detect the expression of ras gene (K-ras, Hs-ras) in 40 cases of laryngeal squamous cell carcinoma Mutation at codon 12 and HPV16, 18 infection. 3 Results In 40 specimens of HPV16-positive in 14 cases (35%), HPV18-positive in 4 cases (10%). There were 9 cases of ras gene mutation, including 6 cases of N-ras, 2 cases of K-ras and 1 case of H-ras. The mutation rate of ras gene in HPV16 positive and negative specimens was 11.11% (2/18) and 31.82% (7/2 2), respectively. There was no significant difference between the two (χ2 = 1.3917, P> 0.05). 4 Conclusion HPV infection and ras gene mutations in the occurrence and development of laryngeal squamous cell carcinoma play an important role.
