目的检测TLR3在正常子宫内膜、子宫内膜异位症(内异症)患者在位和异位内膜中的表达情况,探讨TLR3在内异症发病中的作用。方法选择内异症患者的卵巢异位内膜45例(异位内膜组)和相对应的在位子宫内膜32例(在位内膜组),对照组为32例非内异症患者子宫内膜组织。采用免疫组化方法 (EnVision二步法)检测各组内膜组织中TLR3的表达情况,以及TLR3在内异症不同临床分期的异位内膜组织中的表达差异。结果内异症患者异位内膜、在位内膜及正常对照组子宫内膜中,TLR3蛋白均有不同程度表达,主要表达于子宫内膜腺上皮和腔上皮。TLR3蛋白在内异症患者异位内膜、在位内膜的表达均低于正常对照组子宫内膜,其差异有统计学意义(P<0.01),其中表达最低的是异位内膜,但与在位内膜的差异无统计学意义(P>0.05)。TLR3蛋白在Ⅰ~Ⅱ期内异症患者异位内膜的表达与Ⅲ~Ⅳ期患者异位内膜的表达比较,其差异无统计学意义(P>0.05)。结论在位及异位内膜组织中TLR3低表达可能参与了内异症发病,但与内异症的临床分期无关。
Objective To detect the expression of TLR3 in eutopic and ectopic endometrium of patients with endometriosis and endometriosis (endometriosis) and to explore the role of TLR3 in the pathogenesis of endometriosis. Methods 45 cases of endometriosis (endometriosis group) and 45 cases of endometriosis (endometrium group) were selected for endometriosis in endometriosis group and 32 cases of non-endometriosis control group Endometrial tissue. Immunohistochemistry (EnVision two-step method) was used to detect the expression of TLR3 in each group and the difference of TLR3 expression in ectopic endometrium with different clinical stages. Results In endometriosis and eutopic endometrium of endometriosis patients, TLR3 protein was expressed to varying degrees, which was mainly expressed in endometrial glandular epithelium and luminal epithelium. The expression of TLR3 protein in endometriosis and eutopic endometrium of endometriosis patients was lower than that of normal control group (P <0.01), and the lowest expression of TLR3 protein was ectopic endometrium, However, there was no significant difference with eutopic endometrium (P> 0.05). The expression of TLR3 protein in ectopic endometrium of patients with stage Ⅰ-Ⅱ endometriosis and ectopic endometrium of stage Ⅲ ~ Ⅳ patients showed no significant difference (P> 0.05). Conclusions The low expression of TLR3 in eutopic and ectopic endometrium may be involved in the pathogenesis of endometriosis, but not with the clinical stage of endometriosis.
