目的:检测风湿性心脏病心房颤动患者心房组织中瞬时受体电位通道亚族M7(TRPM7)的mRNA及蛋白水平,探讨TRPM7与房颤心房纤维化的关系。方法:19例风湿性心脏病患者于换瓣术中获取约200 mg的右心房肌组织,分为窦律(SR)组9例和房颤(AF)组10例。HE病理染色观察心肌细胞横截面积,Masson染色及心肌纤维化标志物观察心房纤维化,Western blotting和实时定量PCR测定心房肌组织中TRPM7的mRNA及蛋白水平。结果:与SR组比较,心肌细胞横截面积、胶原容积分数、心肌纤维化标志物CTGF、CollagenⅠ和CollagenⅢ的mRNA水平、TRPM7的mRNA及蛋白水平在AF组显著增加,差异具有统计学意义(P<0.05)。结论:风湿性心脏病房颤患者心肌组织中TRPM7的表达增加,可能通过促心房纤维化,参与房颤的发生与维持。
OBJECTIVE: To detect mRNA and protein levels of TRPM7 in atrial tissue of atrial fibrillation patients with rheumatic heart disease and to investigate the relationship between TRPM7 and atrial fibrillation in atrial fibrillation. Methods: Nineteen patients with rheumatic heart disease received right atrial muscle tissue of about 200 mg during valve replacement surgery. They were divided into sinus rhythm group (SR) group (n = 9) and atrial fibrillation group (n = 10). HE staining, myocardial cell cross-sectional area, Masson staining and myocardial fibrosis markers observed atrial fibrosis, Western blotting and real-time quantitative PCR were used to detect TRPM7 mRNA and protein levels in atrial myocardium. RESULTS: Compared with SR group, the mRNA and protein levels of TRPM7 in myocardium were significantly increased in AF group (P <0.05). The cross-sectional area of myocardial cells, collagen volume fraction, mRNA of CTGF, Collagen Ⅰ and Collagen Ⅲ were significantly increased <0.05). Conclusion: The expression of TRPM7 in myocardium of atrial fibrillation patients with rheumatic heart disease is increased, which may be involved in the occurrence and maintenance of atrial fibrillation through promoting atrial fibrosis.
