目的 :分析抗磷脂综合征患者抗膜突蛋白抗体表达情况,并探讨抗膜突蛋白抗体在抗磷脂抗体相关性血栓发生中可能的作用。方法:采用酶联免疫吸附试验检测70例抗磷脂综合征患者(包括50例抗磷脂抗体相关性血栓患者和20例抗磷脂抗体相关性反复流产者)及100名正常对照者的血清抗膜突蛋白抗体及其他自身抗体的阳性率。制备鼠源性单克隆抗膜突蛋白抗体,加入正常人富血小板血浆中,检测其诱导血小板聚集的能力;同时加入单克隆抗体和膜突蛋白,检测血小板聚集抑制率。结果:在50例抗磷脂抗体相关性血栓患者和20例抗磷脂抗体相关性反复流产患者中,抗N端膜突蛋白抗体阳性率分别为76%和65%,显著高于其他自身抗体(抗C端膜突蛋白抗体、抗血小板抗体、抗心磷脂抗体、抗β2糖蛋白Ⅰ抗体)的阳性率。鼠源性单克隆抗N端膜突蛋白抗体有诱导血小板聚集的作用,而该作用可被N端膜突蛋白抑制,但不能被二磷酸腺苷激活途径抑制物精氨酸-甘氨酸-天冬氨酸-丝氨酸四肽所抑制。结论:大部分抗磷脂抗体综合征患者存在抗N端膜突蛋白抗体阳性,且该抗体的产生可能与抗磷脂综合征患者血栓形成的发病机制相关。抗膜突蛋白抗体在诊断和治疗抗磷脂综合征中的应用值得进一步探索。
OBJECTIVE: To analyze the anti-leukin antibody expression in patients with antiphospholipid syndrome and to explore the possible role of anti-leukin antibodies in anti-phospholipid antibody-related thrombosis. Methods: Serum anti-meningioma was detected by enzyme linked immunosorbent assay in 70 patients with antiphospholipid syndrome (including 50 patients with antiphospholipid antibody-associated thrombosis and 20 patients with recurrent anti-phospholipid antibody abortion) and 100 healthy controls Positive rate of protein antibodies and other autoantibodies. Murine monoclonal anti-N-terminal actin antibody has the effect of inducing platelet aggregation, and this effect can be inhibited by N-terminal ubiquitin but not by the AMP-AMP inhibitor arginine-glycine-aspartate Amino-serine tetrapeptide. Conclusion: Most anti-phospholipid antibody syndrome patients have anti-N-terminal anti-tubulin antibody positive, and the antibody production may be related to the pathogenesis of thrombosis in patients with antiphospholipid syndrome. Anti-idiotoxin antibodies in the diagnosis and treatment of anti-phospholipid syndrome should be further explored.
