目的:探讨艾灸对肾阳虚模型大鼠肾上腺组织形态结构的影响。方法:SD雄性大鼠40只,随机分为空白组、生理盐水组、模型组、艾灸组,每组10只。空白组大鼠正常饲养,不予干预;生理盐水组用0.9%氯化钠后肢肌肉注射,0.3 m L/d,连续28 d;模型组与艾灸组每天用氢化可的松注射液按2.5 mg/100 g后肢肌肉注射,连续14 d以制备肾阳虚模型,造模成功后,两组每天继续予以同剂量氢化可的松注射以维持肾阳虚证,艾灸组同时予关元、肾俞穴隔日交替艾灸治疗,其他3组大鼠同样抓取、捆绑,连续14 d,取材,计算各组大鼠肾上腺系数,在显微镜下观察肾上腺组织形态结构。结果:运用激素造模14 d后,模型组和艾灸组大鼠体质量增长幅度不及空白组明显(P<0.05);治疗14 d后,与模型组比较,艾灸组大鼠体质量增长幅度明显上升(P<0.05),但不及空白组明显(P<0.05);模型组大鼠肾上腺质量及系数较空白组均显著下降(P<0.01),而艾灸组较模型组均明显增加(P<0.01),但仍显著低于空白组(P<0.01);模型组大鼠的肾上腺形态结构有明显损害,肾上腺皮质排列不规则,结构不清晰,皮质变薄、髓质稍增厚。而艾灸组大鼠的肾上腺形态结构损害的程度较模型组有一定程度的减轻。结论:艾灸关元、肾俞穴能够改善肾阳虚模型大鼠阳虚症状,使大鼠体质量增长幅度上升,减轻其肾上腺形态结构损害,增加肾上腺重量及系数。
Objective: To investigate the effects of moxibustion on the morphology of adrenal gland in rats with kidney yang deficiency. Methods: Forty SD male rats were randomly divided into blank group, normal saline group, model group and moxibustion group, with 10 rats in each group. Rats in the blank group were maintained normally without intervention; saline group was intramuscularly injected with 0.9% sodium chloride hindlimb at a dose of 0.3 m L / d for 28 days; model group and moxibustion group were treated with hydrocortisone injection 2.5 mg / 100 g hind limb muscle injection for 14 days to prepare the model of kidney yang deficiency. After the model was established successfully, both groups continued to receive the same dose of hydrocortisone injection daily to maintain the deficiency of kidney-yang, while the moxibustion group was given Guan Yuan, Shenshu acupoints were alternately treated with moxibustion every other day. The other three groups of rats were also grasped and tied for 14 consecutive days. The adrenal coefficient of each group was calculated and the morphology of adrenal tissue was observed under microscope. Results: The body weight of rats in model group and moxibustion group was less than that in blank group after 14-day hormone administration (P <0.05). After 14 days of treatment, compared with model group, the body weight of rats in moxibustion group increased (P <0.05), but not in the blank group (P <0.05). The adrenal gland quality and the coefficient of the model group were significantly decreased compared with the blank group (P <0.01), while the moxibustion group was significantly increased compared with the model group (P <0.01), but still significantly lower than that of the blank group (P <0.01). The morphology of the adrenal gland in the model group was significantly impaired, the adrenal cortex arranged irregularly, the structure was not clear, the cortex became thin and the medulla slightly thicken . However, the degree of adrenal morphological damage in the moxibustion group was relieved to a certain extent compared with the model group. Conclusion: Moxibustion Guanyuan and Shenshu can ameliorate symptoms of yang deficiency in rats with deficiency of kidney - yang, increase the body weight of rats and reduce the damage of adrenal morphology and structure, and increase the weight and coefficient of adrenal glands.
