目的:研究“武当二号金银花”和“武当三号金银花”中主要成分的含量。方法:采用高效液相色谱法-二极管阵列检测器测定绿原酸、芦丁和木犀草苷的含量。测定绿原酸的流动相为乙腈一0.4%磷酸溶液(15:85),色谱柱为Phenomenex Luna-C18(250×4.60 mm,5μm),流速为1.0 m L·min-1,检测波长为327 nm;测定芦丁和木犀草苷流动相为乙腈-0.5%冰醋酸溶液(梯度洗脱),色谱柱为Fortis Xi-Phenyl(250×4.60 mm,5μm),检测波长为354 nm。柱温:30℃。结果:绿原酸、芦丁、木犀草苷在各自测定的范围内均呈良好的线性关系(r≥0.9994),平均回收率为99.5%~99.7%。结论:该方法准确、灵敏度高、重复性好,可用来测定“武当二号金银花”和“武当三号金银花”中绿原酸、芦丁和木犀草苷的含量。
Objective: To study the content of major components in “Wudang II honeysuckle” and “Wudang No. 3 honeysuckle”. Methods: The contents of chlorogenic acid, rutin and luteolin were determined by high performance liquid chromatography-diode array detector. The mobile phase of chlorogenic acid was acetonitrile 0.4% phosphoric acid solution (15:85). The mobile phase consisted of Phenomenex Luna-C18 (250 × 4.60 mm, 5 μm) with a flow rate of 1.0 mL · min-1 and a detection wavelength of 327 nm. The mobile phase of rutin and luteolin was acetonitrile-0.5% glacial acetic acid (gradient elution). The detection wavelength was 354 nm at Fortis Xi-Phenyl (250 × 4.60 mm, 5 μm). Column temperature: 30 ℃. Results: Chlorogenic acid, rutin and luteolin showed a good linear relationship (r≥0.9994) within the range of their determination. The average recoveries were between 99.5% and 99.7%. Conclusion: The method is accurate, sensitive and reproducible. It can be used to determine the contents of chlorogenic acid, rutin and luteolin in “Wudang No.2 Honeysuckle” and "Wudang No.
