目的描述云南省2005年急性弛缓性麻痹(acute flaccid paralysis,AFP)病例中非脊髓灰质炎肠道病毒(non-polio enterovirus,NPEV)的带毒情况和新发现的新型肠道病毒90型(newer enterovirus 90,EV90)的基因特征。方法 2005年云南省共报告232例AFP病例,共采集到<15岁AFP病例222例的双份便标本。对222例AFP标本进行病毒分离,对分离到的NPEV进行VP1基因序列测定。用MEGA5.0软件进行进化树分析和作图。进化树用临位相接法(neighbor joining method)作成,所用模式为Kimura二参数置换法(Kimura 2-parameter substitution model)。用1 000个pseudoreplicate datasets进行进化树bootstrap统计学分析。结果 222例患者中共检测脊髓灰质炎病毒(poliovirus,PV)10株(10/222,4.5%),均为疫苗株,未发现脊髓灰质炎病毒野病毒(wild poliovirus,WPV);检测到NPEV17株(17/222,7.7%),其中4株为人类肠道病毒A组(4个血清型,其中1株为新型病毒EV90),云南株EV90与山东3株之间的核苷酸(nt)同源性为95.7%~96.5%(差异率为3.5%~4.3%),氨基酸(aa)同源性为98.5%~99.5%(差异率为0.5%~1.5%),说明云南株更接近山东株03446-SD-CHN-2003(GQ253428),两地毒株差异不大。12株为人类肠道病毒B组(9个血清型),1株为人类肠道病毒C组(1个血清型,为新型病毒EV96),未分离到人类肠道病毒D组病毒。结论 2005年云南省AFP病例中NPEV携带率与往年持平。新型病毒EV90基因进化分析结果表明EV90病毒存在基因多样性特点。
Objective To describe the virulence of non-polio enterovirus (NPEV) in Yunnan Province in 2005 with acute flaccid paralysis (AFP) and the newly discovered newer enterovirus type 90 enterovirus 90, EV90). Methods A total of 232 cases of AFP were reported in Yunnan Province in 2005. A total of 222 cases of AFP cases were collected. 222 AFP samples were isolated for virus isolation, and NP1V was sequenced. Phylogenetic analysis and mapping using MEGA 5.0 software. The evolutionary tree was created using the neighbor joining method using the Kimura 2-parameter substitution model. Phylogenetic tree bootstrap statistical analysis with 1000 pseudoreplicate datasets. Results A total of 10 strains (10 / 222,4.5%) of poliovirus (PV) were detected in 222 patients. All of them were vaccine strains and no wild poliovirus (WPV) was found. NPEV17 (17 / 222,7.7%), of which 4 were human enterovirus group A (4 serotypes, of which 1 was novel virus EV90), and the nucleotide (nt) The homology was 95.7% ~ 96.5% (the rate of difference was 3.5% ~ 4.3%), and the homology of amino acids was 98.5% ~ 99.5% (the rate of difference was 0.5% ~ 1.5%), indicating that Yunnan strain was closer to Shandong Strain 03446-SD-CHN-2003 (GQ253428), there was no significant difference between the two strains. 12 strains of human enterovirus B (9 serotypes), 1 strain of human enterovirus C (one serotype, a new virus EV96), human enterovirus D virus was not isolated. Conclusion The incidence of NPEV in AFP cases in Yunnan Province in 2005 was the same as in previous years. The result of evolutionary analysis of novel EV90 gene showed that EV90 virus has the characteristics of gene diversity.
