为了观察枸杞子提取液(LBA)对RCS大鼠遗传性视网膜色素变性的治疗作用,将出生后的24只RCS大鼠,随机分为对照组和实验组,每组12只。在实验组RCS大鼠出生后10 d开始喂食1 mg/kg LBA,对照组正常饲养。对照组和实验组RCS大鼠分别在出生后25、35、45 d处死,应用HE染色和TUNEL检测观察视网膜感光细胞的坏死和凋亡,并应用免疫组化的方法检测Caspase-2的表达。结果显示:RCS大鼠实验组25 d和35 d时TUNEL检测阳性细胞数目与对照组相比明显减少(P<0.05);实验组和对照组大鼠生后25 d和50 d时Caspase-2在节细胞层和内核层均有表达,但在25 d时实验组比对照组明显减少(P<0.05)。上述结果提示:在RCS大鼠视网膜变性的早期,LBA通过抑制细胞的凋亡对神经元起保护作用,可以使感光细胞得到更多存留,Caspase-2可促使视网膜变性,LBA则可通过抑制其表达对早期变性的神经元发挥保护作用。
In order to observe the therapeutic effect of wolfberry fruit extract (LBA) on hereditary retinitis pigmentosa in RCS rats, 24 RCS rats after birth were randomly divided into control group and experimental group, 12 in each group. In the experimental group RCS rats were fed 1 mg/kg LBA 10 days after birth and the control group was fed normally. RCS rats in the control and experimental groups were sacrificed at 25, 35, and 45 days after birth respectively. HE staining and TUNEL assay were used to observe the necrosis and apoptosis of photoreceptor cells. The expression of Caspase-2 was detected by immunohistochemistry. The results showed that the number of TUNEL positive cells at the 25th and 35th day of the RCS rat experimental group was significantly lower than that of the control group (P<0.05); Caspase-2 was observed at 25 days and 50 days after birth in the experimental and control groups. It was expressed in both the ganglion cell layer and the inner nuclear layer, but at the 25th day, the experimental group was significantly less than the control group (P<0.05). These results suggest that in the early stage of retinal degeneration in RCS rats, LBA protects neurons by inhibiting cell apoptosis, which allows more photoreceptor cells to persist. Caspase-2 can promote retinal degeneration and LBA can inhibit it. Expression plays a protective role against early degenerating neurons.
