本实验室自从采用液体石腊代替BayolF,用无水羊毛脂代替ArlacelA.以制备油水乳剂作为抗原辅助剂以来,经过数年应用证明可以作为Freund氏油水乳剂的代用品。但所制的油水乳剂不十分稳定,置冰箱(4℃)保存1～2周,油水卽分开,不复是乳剂的状态。经多次改进制备的方法,现已可制成极稳定的乳剂,保存4℃冰箱中数月不分离。取液体石腊85分,加分枝杆菌(每毫升乳剂5mg湿菌)研磨半小时,再加等体积抗原液,再研磨半小时,再加无水羊毛脂15分,继续研磨2小时。研磨系用10毫升注射器去针头,将混合物抽入排出。然后置4℃冰箱内保存24小时,次日再研磨1小时。又置4℃冰箱内
Our laboratory has used liquid paraffin instead of BayolF and anhydrous lanolin instead of Arlacel A for many years as a substitute for Freund’s oil and water emulsion since it was used as an antigen adjuvant. However, the prepared oil and water emulsion is not very stable, set the refrigerator (4 ℃) for 1 to 2 weeks, separate the oil and water, not the emulsion state. After several improvements to the preparation method, it has been made into a very stable emulsion, stored at 4 ℃ refrigerator for several months without separation. Take liquid paraffin 85 points, plus Mycobacterium (per milligram emulsion 5mg wet bacteria) ground for half an hour, plus an equal volume of antigen solution, and then grinding for half an hour, plus anhydrous lanolin 15 minutes, continue grinding for 2 hours. The mill system was deaerated with a 10 ml syringe and the mixture was aspirated and drained. Then set at 4 ℃ refrigerator for 24 hours, then grind the next day for 1 hour. Also set 4 ℃ refrigerator
