目的研究大蒜油对二乙基亚硝胺(N-nitrosodiethylamine,NDEA)诱发大鼠肝癌的抑制作用。方法 60只雄性Wistar大鼠随机分为4组:正常对照组、NDEA模型组、大蒜油低、高剂量处理组,每组15只。NDEA模型组和大蒜油处理组均灌胃NDEA(10 mg/kg),2个大蒜油处理组同时给予不同浓度的大蒜油灌胃(20和40 mg/kg),正常对照组给予等体积的玉米油灌胃。观察大鼠体重、肝重、肝脏系数、癌结节数和癌结节发生率的变化,同是对肝脏进行病理组织学检查;并检测血清中丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、碱性磷酸酶(ALP)、γ-谷氨酰转肽酶(γ-GT)的活性和甲胎蛋白(AFP)的含量。结果与正常对照组相比,NDEA模型组大鼠体重明显降低,肝重和肝脏系数明显增加,血清ALT、AST、ALP和γ-GT的活性和AFP的含量均明显升高(P<0.01);与NDEA模型组相比,大蒜油处理组大鼠各项指标明显好转(P<0.01或P<0.05)。病理结果显示,与NDEA模型组相比,大蒜油低、高剂量组癌结节数和发生率明显降低,且癌细胞分化程度较高。结论大蒜油对NDEA诱发大鼠肝癌形成具有明显的抑制作用。
Objective To study the inhibitory effect of garlic oil on liver cancer induced by N-nitrosodiethylamine (NDEA) in rats. Methods Sixty male Wistar rats were randomly divided into 4 groups: normal control group, NDEA model group, low and high garlic oil treatment group, 15 rats in each group. NDEA model group and garlic oil treatment group were fed with NDEA (10 mg / kg), two garlic oil treatment groups were given different concentrations of garlic oil (20 and 40 mg / kg), the normal control group were given equal volume Corn oil gavage. The changes of body weight, liver weight, liver coefficient, the number of nodules and the incidence of cancer nodules were observed, and the pathological changes of the liver were also examined. Serum levels of alanine aminotransferase (ALT), aspartate Aminotransferase (AST), alkaline phosphatase (ALP), γ-glutamyl transpeptidase (γ-GT) activity and alpha-fetoprotein (AFP) Results Compared with the normal control group, the body weight of rats in NDEA model group decreased significantly, the liver weight and liver coefficient increased significantly, the activities of serum ALT, AST, ALP and γ-GT and the content of AFP increased significantly (P <0.01) Compared with NDEA model group, the indexes of garlic oil treatment group improved significantly (P <0.01 or P <0.05). The pathological results showed that compared with the NDEA model group, the number and incidence of cancer nodules in the low and high dose garlic oil groups were significantly decreased, and the degree of cancer cell differentiation was high. Conclusion Garlic oil has obvious inhibitory effect on NDEA-induced hepatocarcinogenesis in rats.
