目的验证中华大蟾蜍具有抗结核活性,筛选抗结核活性成分。方法对中华大蟾蜍水提物进行葡聚糖凝胶分子筛分离和反相高效液相层析法(RPLC)分离,并采用微孔板Alamar blue法将分离所得12组分进行体外抑菌实验研究。结果第1、6、10组分表现出对供试结核分枝杆菌的明显抑制作用,其最低抑菌浓度(minimal inhibitory concentration,MIC)分别为339.98mg/ml、20.00mg/ml、2.95mg/ml。结论中华大蟾蜍含有抑制结核分枝杆菌的活性成分,该有效成分分子量接近14KD,并表现出弱极性的特征。
Objective To verify the anti-tuberculosis activity of Chinese cockroach and screen out the anti-tuberculous active ingredients. Methods The Selaginella sinensis water extract was separated by Sephadex gel molecular sieve and separated by reversed-phase high performance liquid chromatography (RPLC), and the 12 components isolated by Alamar blue method were tested in vitro. . Results The 1st, 6th and 10th components showed significant inhibitory effect on the tested Mycobacterium tuberculosis. The minimal inhibitory concentration (MIC) was 339.98mg/ml, 20.00mg/ml, 2.95mg/ respectively. Ml. Conclusion The Chinese cockroach contains active ingredients that inhibit Mycobacterium tuberculosis. The molecular weight of the active ingredient is close to 14KD and shows a weak polarity.
