血管内皮生长因子(VEGF)是内皮细胞的特异性致裂原,在血管发生和血管生成过程中占有重要地位.近年来发现,VEGF对胚胎植入也具有一定作用,但有关VEGF与植入中标记分子MMPs之间的关系,迄今未见报道.因此,研究目的是阐明VEGF在小鼠胚胎植入过程中对MMPs的影响.半定量RT-PCR的结果表明,VEGF抗体可显著抑制小鼠子宫MMP-2和-9 mRNA的表达;将小鼠胚泡培养于FN铺碟的培养液中,同时用不同浓度的VEGF抗体孵育胚泡,通过明胶酶谱分析检测发现,0.1 和1μg/mL的 VEGF抗体能明显降低胚泡分泌的MMP-2和-9水解明胶活性的能力.以上结果提示,VEGF抗体可干扰子宫内膜或胚泡MMP2和-9的基因表达和蛋白分泌,使滋养层细胞向子宫内膜的侵入能力或者子官内膜接受胚泡的能力降低,影响胚胎植入过程.
