杜仲成熟干种子经GA3溶液浸泡后,去掉种胚,切取胚根端胚乳接种在愈伤组织诱导培养基上,5~7d后多数胚乳顶端局部膨大。将膨大部分切下转入成分相同的新鲜培养基上,40~50d后形成白色柔软的初生愈伤组织。初生愈伤组织经改造后变为绿色致密具分化能力的愈伤组织,转入分化培养基后出现器官分化,唯正常茎芽分化频率很低,且十分细弱,只有经壮苗培养后才能诱导生根。目前已获得胚乳试管苗20余株。其无性系中部分植株已在温室内移栽成活。
Eucommia mature seeds were soaked in GA3 solution, the embryos were removed and the radicle endosperm was inoculated on the callus induction medium. Most of the endosperm enlarged locally after 5-7 days. The swollen part of the cut into the same composition of fresh medium, 40 ~ 50d after the formation of white soft primary callus. After primary transformation, the callus transformed into green compact callus with differentiation ability. After differentiation into differentiation medium, organ differentiation occurred. However, the frequency of normal stem bud differentiation was very low and very weak. Only after strong seedling culture did it induce root. So far, more than 20 endosperm test tube seedlings have been obtained. Some of its clones have transplanted alive in the greenhouse.
