目的探讨大鼠海马区及皮层EphA4表达在慢性脑缺血的不同时间点的动态变化。方法大鼠随机分为正常组、假手术组、15d、30d和60d模型组.采用双侧颈总动脉永久结扎建立慢性脑缺血模型;Morris水迷宫试验检测行为学变化;免疫组化染色检测EphA4的表达变化。结果EphA4免疫反应阳性细胞数模型组比假手术组均增多(P<0.05),与行为学改善基本相符。结论慢性脑缺血造成EphA4表达持续升高可能是成年哺乳动物损伤后神经再生障碍的重要原因之一。
Objective To investigate the dynamic changes of EphA4 expression in rat hippocampus and cortex at different time points after chronic cerebral ischemia. Methods The rats were randomly divided into normal group, sham operation group, 15d, 30d and 60d group, chronic cerebral ischemia model was established by bilateral common carotid artery ligation, Morris water maze test was used to detect behavioral changes, immunohistochemical staining EphA4 expression changes. Results The number of EphA4 immunoreactive positive cells in model group was higher than that in sham operation group (P <0.05), which was consistent with the improvement of behavior. Conclusion The persistent increase of EphA4 expression induced by chronic cerebral ischemia may be one of the important reasons for the impaired neural regeneration in adult mammals.
