目的: 用高效液相色谱法测定兔血清中琥乙红霉素的浓度。方法: 采用YWG- C18 色谱分析柱(4-6 m m×150 m m ,10 μm), 岛津SPD- 6AV 紫外检测器, 甲醇- 0-02 mol·L- 1 - 醋酸铵水溶液(70∶30 , 用氨水调pH7-3) 为流动相, 红霉素作内标。血液样品在碱性条件下用二氯甲烷提取纯化后进样, 在210 nm 波长处检测。结果: 琥乙红霉素线性范围为0-25~12 mg·L- 1 (r= 0-998 9), 最低检测量0-02 μg, 最低检出浓度0-05 mg·L-1 , 平均方法回收率96-26% 。精密度考察琥乙红霉素日内、日间相对标准偏差均小于6% 。结论: 本方法快速、准确、重复性好。
Objective: To determine the concentration of erythromycin ethylsuccinate in rabbit serum by high performance liquid chromatography. Methods: The YWG-C18 column (4-6 m × 150 m m, 10 μm), Shimadzu SPD-6AV UV detector, methanol-0-02 mol·L -1 ammonium acetate aqueous solution 30, adjust pH7-3 with ammonia) as the mobile phase, erythromycin as internal standard. The blood samples were extracted with dichloromethane after purification under alkaline conditions and injected at 210 nm. Results: The linear range of erythromycin ethylsuccinate was 0-25 ~ 12 mg · L -1 (r = 0-998 9), the lowest detection limit was 0-02 μg, the lowest detection limit was 0-05 mg · L -1, The average recovery rate of 96-26%. The intra-day and inter-day relative standard deviations (RSDs) of erythromycin ethylsuccinate were all less than 6%. Conclusion: The method is rapid, accurate and reproducible.
