目的 :在常规条件下培养成年猴的许旺细胞。方法 :用神经结扎术分别结扎年龄为 3 ,9,13岁的 3只雄性恒河猴的腓肠神经 ,1周后 ,取结扎处远段的神经进行植块培养、纯化 ,并测定其在聚酯纤维上的迁移速度。结果 :许旺细胞从神经段中迁出的时间 ,3岁猴为 3d ;9岁猴为 5d ;13岁猴为 7d。许旺细胞迁出的数量和猴的年龄成反比。 3只猴的许旺细胞均具有较良好的迁移活力。许旺细胞抗S 10 0蛋白抗体染色阳性。 3只猴的神经段中均有植块无许旺细胞迁移 ,但有髓鞘泡存在。结论 :结扎神经使其发生瓦勒变性 ,在常规条件下经植块培养、纯化 ,能够获得可用于移植的成年猴的许旺细胞。
OBJECTIVE: To cultivate Schwann cells of adult monkeys under routine conditions. Methods: The sural nerve of 3 male Rhesus macaques aged 3, 9 and 13 years old were ligated by nerve ligation. One week later, the nerves in the distal part of the ligation were harvested for explant culture and purified. Migration speed on polyester fiber. Results: The time of Schwann cells migrating from the nerve segment was 3 days for 3-year-old monkeys, 5-year-old for 9-year-old monkeys and 7 days for 13-year-old monkeys. The number of Schwann cells migrated is inversely proportional to the age of the monkeys. Schwann cells of 3 monkeys had better migration activity. Schwann cells were stained positive for anti-S 10 0 antibody. There were no Schwann cells migrating in the nerve segments of 3 monkeys, but the myelinated vesicles existed. CONCLUSION: The nerve is ligated to transform Wallerian degeneration. After culturing and purifying it under normal conditions, the Schwann cells can be obtained from adult monkeys which can be used for transplantation.
