目的:观察COX-2与Survivin在SMMC-7721细胞中的表达情况及COX-2抑制剂NS-398对二者表达的影响,探讨NS-398的抗肿瘤作用机制。方法:以不同浓度的NS-398(0、25、50、75、100、150μmol/L)处理SMMC-7721细胞24、48、72小时,应用qRT-PCR检测COX-2、Survivin mRNA的表达,应用Western blot检测COX-2、Survivin蛋白的表达,应用FCM鉴定COX-2、Survivin蛋白表达。结果:COX-2与Survivin在正常生长的SMMC-7721细胞中均有较高表达,NS-398处理后,COX-2、Survivin基因和蛋白表达均显著减少,即与NS-398的作用剂量及时间呈反比。结论:NS-398可能是通过降低COX-2与Survivin的表达来实现改变细胞周期、诱导凋亡,从而抑制肿瘤的生长。
OBJECTIVE: To observe the expression of COX-2 and Survivin in SMMC-7721 cells and the effect of COX-2 inhibitor NS-398 on the expression of COX-2 and anti-tumor effect of NS-398. Methods: SMMC-7721 cells were treated with different concentration of NS-398 (0, 25, 50, 75, 100 and 150μmol / L) for 24,48,72 hours. The expressions of COX-2 and Survivin mRNA were detected by qRT- The expression of COX-2 and Survivin protein was detected by Western blot. The expression of COX-2 and Survivin protein was detected by FCM. Results: COX-2 and Survivin were highly expressed in normal SMMC-7721 cells. After treatment with NS-398, the expression of COX-2 and Survivin gene and protein were significantly decreased, Time is inversely proportional. CONCLUSIONS: NS-398 may inhibit the growth of tumor by reducing the expression of COX-2 and Survivin to change the cell cycle and induce apoptosis.
