目的观察异体角膜体外对人外周血T细胞及其亚群CD28分子表达的调节。方法应用异体角膜与外周血淋巴细胞体外共同培养模型和流式细胞分析技术(fluoresceinactivatedcelsorter,FACS)。结果对照组T细胞CD28表达为21.3%;受角膜或沸波醇酯(phorbol12,13—dibutyrate,PDB)激活后,T细胞CD28表达分别为52.4%和81.4%;受角膜激活后,CD4和CD8T淋巴细胞CD28表达分别为63.1%和52.3%。结论角膜组织体外能够刺激人外周血T细胞CD28表达和T细胞活化,CD4T细胞比CD8T细胞活化更明显。
Objective To observe the regulation of the expression of CD28 on T cells and their subpopulations in human peripheral blood in vitro. Methods The in vitro co-culture model of allogeneic cornea and peripheral blood lymphocytes and the flow cytometry (FACS) were used. Results The CD28 expression of T cells in control group was 21.3%. After activated by cornea or phorbol 12,13-dibutyrate (PDB), CD28 expression of T cells was 52.4% and 81.4% After activation, the CD28 expression of CD4 and CD8 T lymphocytes were 63.1% and 52.3%, respectively. Conclusions Corneal tissue can stimulate CD28 expression and T cell activation of T lymphocytes in vitro, and CD4 T cells activate more than CD8 T cells.
