为探讨四逆汤抗实验性动脉粥样硬化的作用及其可能机制 ,运用图像分析法测定主动脉内膜脂质斑块面积百分比 ,脂代谢指标分别运用酶动力学法、一步法、公式计算法、免疫透射比浊法等方法测定 ,血清一氧化氮及血浆内皮素分别用酶法及放射免疫法检测 ,血浆超氧化物歧化酶与丙二醛分别运用微量快速测定法、改良的八木国夫法测定。结果发现 ,四逆汤可明显缩小主动脉内膜脂质斑块面积 ,降低血清总胆固醇、甘油三酯、低密度脂蛋白胆固醇、载脂蛋白B及血浆内皮素和丙二醛浓度 ,提高血清一氧化氮水平、载脂蛋白A含量与血浆超氧化物歧化酶活性 ,四逆汤高剂量组效果为佳。结果提示 ,四逆汤具有较好的抗实验性动脉粥样硬化作用 ,其作用机制与调节脂代谢、抗脂质过氧化损伤及保护血管内皮细胞功能等有关。
To investigate the effect of Sini decoction on experimental atherosclerosis and its possible mechanism, image analysis was used to determine the percentage of aortic intima lipid plaque area. Lipid metabolism was measured by enzyme kinetics, one-step method and formula calculation respectively. Methods such as immunoassay and immunoturbidimetric method were used to measure serum nitric oxide and plasma endothelin by enzymatic method and radioimmunoassay respectively. Plasma superoxide dismutase and malondialdehyde were detected by trace fast assay and improved Yagi Kazuo respectively. Method determination. The results showed that Sini Decoction can significantly reduce the area of aortic intima lipid plaque, reduce serum total cholesterol, triglyceride, low-density lipoprotein cholesterol, apolipoprotein B, plasma endothelin and malondialdehyde concentrations, and increase serum Nitric oxide levels, apolipoprotein A levels, and plasma superoxide dismutase activity were better in the high-dose Sini Tang group. The results suggest that Sini Decoction has good anti-atherosclerotic effects, and its mechanism of action is related to the regulation of lipid metabolism, anti-lipid peroxidation injury, and protection of vascular endothelial cell function.
