目的:研究参麦注射液体外对骨肉瘤MG-63细胞生长的影响及作用机制。方法:体外培养MG-63细胞,以不同浓度的参麦注射液作用于细胞,MTT法检测肿瘤细胞生长抑制率;流式细胞术测细胞凋亡率和细胞周期;ELISA法测凋亡基因Caspase-3、Caspase-9的含量;Bradford法测SOD、MDA含量。结果:参麦注射液能显著抑制MG-63细胞的增殖,呈剂量依赖性;增加MG-63细胞的凋亡率,并诱导出现S细胞周期阻滞;作用MG-63细胞后,Caspase-3和Caspase-9蛋白表达量增加;升高细胞内SOD的活性,降低MDA的含量。结论:参麦注射液抑制人骨肉瘤MG-63细胞增殖的作用机制与诱导细胞凋亡和将细胞周期阻滞于S期有关,其诱导凋亡的机制与Caspase家族,以及增加机体的抗氧化功能有关。
Objective: To study the effect of Shenmai injection on the growth of osteosarcoma MG-63 cells in vitro and its mechanism. Methods: MG-63 cells were cultured in vitro and treated with different doses of Shenmai injection. The growth inhibition rates of tumor cells were detected by MTT assay. The apoptosis rate and cell cycle were measured by flow cytometry. The expressions of apoptosis gene Caspase -3, Caspase-9 content; Bradford test SOD, MDA content. Results: Shenmai injection can significantly inhibit the proliferation of MG-63 cells in a dose-dependent manner; increase the apoptosis rate of MG-63 cells and induce the cell cycle arrest of S-cell; after treated with MG-63 cells, Caspase-3 And Caspase-9 protein expression increased; increase intracellular SOD activity, reduce MDA content. Conclusion: The mechanism of Shenmai injection in inhibiting the proliferation of human osteosarcoma MG-63 cells is related to the induction of apoptosis and cell cycle arrest in S phase. The mechanism of apoptosis induced by Shenmai injection is related to the Caspase family and the anti-oxidant function related.
