目的:探讨肾复宁颗粒治疗慢性肾功能不全(chronic renal failure,CRF)的作用机制。方法:SD大鼠采用右肾切除左肾外动脉分支结扎方法建立CRF模型,相应药物连续治疗8周,观察血清Scr、BUN及肾脏组织中TGF-β1、CTGF的mRNA水平。结果:CRF大鼠经肾复宁颗粒治疗后,血清Scr、BUN及肾组织TGF-β1、CTGF的mRNA水平明显降低,与模型组比较P<0.01。结论:肾复宁颗粒通过抑制TGF-β1mRNA、CTGFmRNA的表达,从而改善CRF大鼠肾功能,达到延缓肾脏纤维化作用,其疗效具有一定剂量正向相关性。
Objective: To investigate the mechanism of Shenfuning granule in treating chronic renal failure (CRF). Methods: CRF models were established by right nephrectomy in the left renal artery. The corresponding drugs were continuously treated for 8 weeks. The levels of mRNA of TGF-β1 and CTGF in the serum of Scr, BUN and kidney were observed. Results: The CRF rats were treated with Shenfuning Granules, and the mRNA levels of Scr, BUN and TGF-β1 and CTGF in renal tissue were significantly lower than those in model group (P <0.01). Conclusion: Shenfoning granule can inhibit the renal function of rats with CRF by inhibiting the expression of TGF-β1mRNA and CTGFmRNA, which can delay the renal fibrosis. The therapeutic effect is dose-dependent.
