目的观察不同培养条件对内皮细胞(EC)纤溶活性的影响。方法用ELISA法测定人脐静脉内皮细胞(HUVEC)培养上清液中的纤溶酶原激活物(t-PA)及I型纤溶酶原灭活剂(PAI-1)总抗原,观察不同代EC分泌t-PA,PAI-1抗原的变化,以及不同血清浓度对两种抗原含量的影响。结果传代和血清均可刺激PAI-1抗原的合成分泌;HUVEC在第3~4代分泌PAI-1抗原量相当;10%血清浓度时PAI-1抗原含量最高。结论传代和不同浓度血清可影响HUVEC分泌PAI-1抗原,但不影响t-PA抗原的分泌。
Objective To observe the effect of different culture conditions on the fibrinolytic activity of endothelial cells. Methods The total antigen of plasminogen activator (t-PA) and type I plasminogen activator (PAI-1) in culture supernatant of human umbilical vein endothelial cells (HUVEC) were determined by ELISA. Generation of EC secretion of t-PA, PAI-1 antigen changes, as well as different serum concentrations of two antigen content. Results Both passage and serum could stimulate the synthesis and secretion of PAI-1 antigen. The amount of PAI-1 antigen secreted by HUVEC was similar at the 3rd to 4th passage, and the highest PAI-1 antigen at 10% serum concentration. Conclusion Passage and different concentrations of serum can affect the secretion of PAI-1 antigen in HUVECs, but does not affect the secretion of t-PA antigen.
