目的:探讨水通道蛋白3(AQP3)在胃腺癌不同分期中的差异表达与淋巴结转移的关系。方法:采用定量PCR、Western blot法对80例不同分期的胃腺癌组织及作为对照的癌旁非肿瘤组织中AQP3 mRNA和蛋白的表达分别进行检测,并与各临床病理分期中淋巴结转移情况进行分析。结果:AQP3在胃腺癌不同分期中的差异表达影响胃癌的淋巴结转移,有淋巴结转移的T1、T2、T3、T4期(UICC,2010)肿瘤组织中AQP3的表达明显高于无淋巴结转移者。结论:胃腺癌肿瘤组织中高表达的AQP3可能对患者淋巴结转移的发生有促进作用,因此AQP3有望成为预测胃腺癌转移和评估预后的重要指标,并可能是胃癌治疗靶点之一。
Objective: To investigate the relationship between differential expression of aquaporin 3 (AQP3) in different stages of gastric adenocarcinoma and lymph node metastasis. Methods: The expression of AQP3 mRNA and protein in 80 cases of gastric adenocarcinoma with different stages and adjacent non-tumor tissues were detected by quantitative PCR and Western blot, respectively. The expression of AQP3 mRNA and protein was analyzed with the clinicopathological data of lymph node metastasis . Results: The differential expression of AQP3 in different stages of gastric adenocarcinoma affected lymph node metastasis. The expression of AQP3 in T1, T2, T3, T4 (UICC, 2010) with lymph node metastasis was significantly higher than that without lymph node metastasis. CONCLUSION: AQP3, which is overexpressed in gastric adenocarcinoma, may play a role in the development of lymph node metastases. Therefore, AQP3 is expected to be an important index for predicting the metastasis and prognosis of gastric adenocarcinoma and may be one of the therapeutic targets for gastric cancer.
