目的探讨兔下颌骨牵张成骨中HIF-1α和c-fos的表达及其相关性。方法建立25只成年健康家兔下颌骨牵张成骨模型,采用免疫组织化学方法,分别在牵张中期、牵张末期、固定期第1、3、5周,检测牵张区新骨组织中HIF-1α和c-fos的分布及表达变化。结果HIF-1α在牵张末期和固定第1周表达为强阳性,c-fos在牵张中、末期和固定第1周表达为强阳性,两者均明显强于固定第3、5周。且均主要表达于成纤维细胞、成骨细胞和新埋入骨基质中的骨细胞中。结论在下颌骨缺损牵张成骨中HIF-1α和c-fos在细胞中的分布及在时间点的表达具有显著的相关性,对骨及血管的生成可能起着重要的生物学作用。
Objective To investigate the expression of HIF-1α and c-fos in rabbit mandibular distraction osteogenesis and its correlation. Methods The mandibular distraction osteogenesis model was established in 25 adult healthy rabbits. Immunohistochemical staining was performed in the mid-stretch, end-stretch and first, third, fifth and fifth week respectively. HIF-1α and c-fos distribution and expression changes. Results The expression of HIF-1α was strongly positive at the end of stretch and at the first week of fixation. The expression of c-fos was strongly positive in the middle, end and first week of stretch, both of which were significantly stronger than those of the first and third week. And were mainly expressed in fibroblasts, osteoblasts and newly embedded osteocytes in bone matrix. Conclusion The distribution of HIF-1α and c-fos in the cells during distraction osteogenesis of mandibular defects has a significant correlation with the expression at the time point, which may play an important biological role in the formation of bone and blood vessels.
