采用气相色谱法测定糖类,具有灵敏、快速、选择性高等优点。但因糖在受热时没有足够的挥发性,必须在气相色谱法分析之前预先制备成易挥发、对热稳定的衍生物。对不同的样品可以制备不同的衍生物,由于糖肟三甲基硅醚衍生物有其特点,对糖浆或非结晶糖样品都可以进行测定,同时可以减少衍生物异构体的生成,使多数单糖都能得到单一的色谱峰。关键的问题是色谱分离D-甘露糖、D-半乳糖和D-葡萄糖情况较差,因而影响定性、定量分析。针对这个问题,我们对制备糖肟三甲基硅醚常规方法作些改进,使糖肟三甲基硅醚衍生物主要以单肟存在,这样三种己糖的构型就不同,就可以得到分离。
