以2个北方粳型恢复系津稻162和津稻18为受体,经农杆菌EHA105/pDSBar1300转化,分别获得77和55个转植株。用0.25%浓度的Basta对转基因在T0代进行了涂布试验,其抗性植株率分别为92%和78%,对除草剂抗性转基因株系做PCR检测,均能扩增出400bp的Bar基因特异条带,证明外源基因已整合到水稻基因组中。对转基因T1代进行潮霉素抗性鉴定,大多数转基因株系表现为3∶1的分离比。
Two northern japonica restorer lines, Jindi 162 and Jindi 18, were transformed with Agrobacterium tumefaciens EHA105 / pDSBar1300 to obtain 77 and 55 transformants respectively. Transgenic plants with 0.25% Basta were tested for coating on T0 generation with resistance rates of 92% and 78%, respectively. PCR assays of the herbicide resistant transgenic lines yielded a 400 bp Bar Gene-specific bands to prove that foreign genes have been integrated into the rice genome. Transgenic T1 generation was hygromycin resistant and most of the transgenic lines showed a segregation ratio of 3: 1.
