目的探讨血管活性肠肽(vasoactive intestinal peptide,VIP)对慢性阻塞性肺疾病(chronic obstructivepulmonary disease,COPD)患者外周血单个核细胞分泌肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)水平的影响。方法收集健康组15例和COPD急性加重期患者15例外周血单个核细胞,并且将COPD急性加重期(acuteepisode of chronic obstructive pulmonary disease,AECOPD)患者的外周血单个核细胞随机分为空白亚组及VIP亚组,分别进行体外培养,应用放射免疫方法测定3组外周血单个核细胞上清液中TNF-α含量的变化。结果对照组细胞上清液中TNF-α含量低于空白亚组(P<0.05);VIP亚组细胞上清液中TNF-α低于空白亚组(P<0.05)。结论 COPD急性发作期患者体内存在严重的炎症反应,VIP能够降低AECOPD患者单个核细胞分泌炎症介质TNF-α分泌,可能是其阻止COPD炎症反应的重要机制之一。
Objective To investigate the effect of vasoactive intestinal peptide (VIP) on the secretion of tumor necrosis factor-α (TNF-α) from peripheral blood mononuclear cells in patients with chronic obstructive pulmonary disease (COPD) Impact. Methods Peripheral blood mononuclear cells were collected from 15 healthy subjects and 15 acute exacerbation COPD patients. Peripheral blood mononuclear cells (PBMCs) were randomly divided into blank subgroups and control subjects with acute epileptic of chronic obstructive pulmonary disease (COPD) VIP subgroups were cultured in vitro, the radioimmunoassay method was used to measure the changes of TNF-αin the peripheral blood mononuclear cell supernatant of the three groups. Results The level of TNF-α in the supernatant of the control group was lower than that of the blank group (P <0.05). The level of TNF-α in the supernatant of VIP group was lower than that of the blank group (P <0.05). Conclusions Severe inflammatory reaction exists in patients with COPD during acute exacerbation. VIP can reduce TNF-α secretion from inflammatory cells in mononuclear cells of AECOPD patients, which may be one of the important mechanisms for preventing COPD inflammatory reaction.
