目的:建立周氏克金岩方中6种黄酮指标成分的快速UV在线检测方法。方法:用HPLC测定39批周氏方提取液中儿茶素、羟基红花黄色素A、金丝桃苷、异槲皮苷、槲皮苷、槲皮素6种黄酮指标成分含量;同时将这39批提取液在200~500 nm进行UV全波长扫描,取每隔1 nm的紫外吸光度。分别将HPLC测得各提取液6指标成分含量与其301个紫外吸光度进行逐步回归,建立6种指标成分含量的UV预测方程,另取8批周氏方提取液分别用HPLC和UV预测法测定其实际含量和预测含量,检验预测结果的可靠性。结果:6种指标成分UV预测方程的相关系数依次为0.935,0.963,0.911,0.956,0.926,0.890。8批验证提取液中各指标成分的实际含量与预测含量结果的相关系数分别为0.946 5,0.976 5,0.907 9,0.981 9,0.911 4,0.896 2;两组值间偏差小;经t检验,两组数据间没有显著性差异。结论:UV预测法结果可靠、准确,方法快速、方便,可用于周氏方生产过程中6种黄酮指标成分的在线检测。
OBJECTIVE: To establish a rapid on-line UV detection method for six flavonoid components in Zhoushi Kejinyan prescription. Methods: Sixteen flavonoid components of catechin, hydroxysafflor yellow A, hyperoside, quercetin and quercetin in 39 batches of Zhou’s extract were determined by HPLC. The 39 batches of extracts were scanned at UV wavelengths from 200 nm to 500 nm with UV absorbance at 1 nm intervals. The HPLC method was used to determine the UV predictive equation of 6 index components by the stepwise regression of 6 components and 301 UV absorbance of each extract. The other 8 batches of Zhou’s extract were determined by HPLC and UV prediction respectively Actual content and predicted content, test the reliability of prediction results. Results: The correlation coefficients of the UV predictive equations of the six kinds of index components were 0.935, 0.963, 0.911, 0.956, 0.926 and 0.890 respectively. The correlation coefficients between the actual content and the predicted content of each index component in the batch were 0.946 5, 0.976 5,0.907 9,0.981 9,0.911 4,0.896 2; the deviation between the two groups was small; by t test, there was no significant difference between the two groups of data. Conclusion: The UV prediction method is reliable and accurate, and the method is rapid and convenient. It can be used for on-line detection of six flavonoid components in the process of production.
