目的探讨骨髓间充质干细胞对肾癌细胞系A498增殖和侵袭能力的影响。方法将骨髓间充质干细胞及人成纤维细胞分别与肾癌A498细胞共培养,利用流式细胞仪测定细胞周期,检测骨髓间充质干细胞及人成纤维细胞对A498细胞增殖能力的影响;Transwell侵袭实验分析肾癌A498细胞体外侵袭能力;qRT-PCR检测共培养后肾癌A498细胞Snail和E-cadherin基因的表达。结果骨髓间充质干细胞可明显抑制A498细胞的增殖及侵袭能力(P<0.05);qRT-PCR结果表明,与骨髓间充质干细胞共培养后,A498细胞中Snail基因表达下调(P<0.05),而E-cadherin基因表达上调(P<0.05)。成纤维细胞对A498细胞所起的作用与骨髓间充质干细胞相反。结论骨髓间充质干细胞体外可抑制A498细胞的增殖和侵袭。
Objective To investigate the effect of bone marrow mesenchymal stem cells on proliferation and invasion of renal cell carcinoma cell line A498. Methods Bone marrow mesenchymal stem cells and human fibroblasts were co-cultured with human renal cell carcinoma cell line A498 respectively. The cell cycle was measured by flow cytometry to detect the effect of bone marrow mesenchymal stem cells and human fibroblasts on the proliferation of A498 cells. Transwell Invasion assay was used to analyze the invasion ability of renal cell carcinoma A498 cells in vitro. QRT-PCR was used to detect the expression of Snail and E-cadherin gene in renal cell carcinoma A498 cells. Results BMSCs could significantly inhibit the proliferation and invasion ability of A498 cells (P <0.05). The results of qRT-PCR showed that the expression of Snail gene was down-regulated in A498 cells after co-cultured with BMSCs (P <0.05) , While E-cadherin gene expression was up-regulated (P <0.05). The effect of fibroblasts on A498 cells is opposite to that of bone marrow mesenchymal stem cells. Conclusion Bone marrow mesenchymal stem cells can inhibit the proliferation and invasion of A498 cells in vitro.
