目的探讨邻苯基苯酚(OPP)的细胞毒性作用及其机制。方法通过体外细胞试验,用OPP作用HepG2细胞,通过彗星试验观察DNA链断裂、测定ROS、GSH水平和抗氧化剂TH干预后ROS、GSH水平变化。结果 OPP能够引起细胞DNA损伤(尾DNA%由空白的0.06增加为800μmol/L OPP的0.98等);细胞内ROS水平增高(P<0.01)和GSH水平降低(P<0.01);TH抗氧化干预效果明显。结论 OPP对HepG2细胞具有遗传毒性,其损伤机制与氧化应激有关。
Objective To investigate the cytotoxicity of o-phenylphenol (OPP) and its mechanism. Methods HepG2 cells were treated with OPP in vitro and the DNA strand breaks were observed by comet assay. The levels of ROS and GSH in serum and ROS, GSH levels were measured after intervention with TH. Results OPP induced DNA damage (the percentage of tail DNA was increased from 0.06 to 0.98 for 800μmol / L OPP, the level of ROS was increased (P <0.01) and the level of GSH was decreased (P <0.01) The effect is obvious. Conclusion OPP is genotoxic to HepG2 cells, and its mechanism is related to oxidative stress.
