目的确定二精方中总多糖的提取工艺并进行含量测定。方法分别采用回流提取法与超声提取法提取枸杞总多糖、黄精总多糖及二精方中总多糖,通过苯酚-硫酸法测定其多糖含量,确定最佳提取工艺。结果标准曲线:Y=45.92-0.030 4,R2=0.999 7;精密度RSD为0.974 4%;重复性RSD为2.953 8%;稳定性RSD为1.766 9%;加样回收率为98.48%。二精方回流提取得率17.546%,含量45.4%;二精方超声提取得率12.915%,含量57.4%;枸杞回流黄精超声提取二精方多糖,含量70.0%;枸杞超声黄精回流提取二精方多糖,含量48.5%。结论二精方多糖最佳提取工艺为枸杞回流提取黄精超声提取,合并提取多糖。
Objective To determine the extraction process of total polysaccharide from Erjing Fang and determine the content of total polysaccharide. Methods The total polysaccharide of Lycium barbarum, total polysaccharides of Polygonatum sibiricum and total polysaccharides of Erjing were extracted by reflux extraction and ultrasonic extraction respectively. The optimum polysaccharide content was determined by phenol-sulfuric acid method. Results Standard curve: Y = 45.92-0.0304, R2 = 0.999 7; RSD of precision was 0.974 4%; RSD of repeatability was 2.953 8%; RSD of stability was 1.766 9%; recovery of sample was 98.48%. The yield of two spermine was 17.546%, the content was 45.4%; the yield of spermine was 12.915% and the content was 57.4% by ultrasonic extraction; Polysaccharides, content of 48.5%. Conclusion The optimum extraction process of polysaccharides from the two spermine polysaccharides by reflux extraction of Polygonatum sibiricum ultrasonic extraction, combined extraction of polysaccharides.
