用改进的Klenk法从狗红细胞中提取和纯化了神经节苷脂GM3,将分离纯化的GM3加入早幼粒白血病细胞系(HL-60)DME/F12培养液中,终浓度为50μM,细胞的增殖明显受到抑制,并按单核-巨噬细胞分化成熟,表现为细胞吞噬功能显著增强,非特异性酯酶活性升高。细胞的形态呈现与分化相一致的变化,比较GM3诱导HL-60细胞分化前后细胞中性糖脂图谱的变化,CDH显著增多是GM3诱导HL-60细胞向单核-巨噬细胞分化的一个重要特征,CMH及CQH亦相应显著增多。
The ganglioside GM3 was extracted and purified from dog erythrocytes using a modified Klenk method. The isolated and purified GM3 was added to the promyelocytic leukemia cell line (HL-60) in DME/F12 medium to a final concentration of 50 μM. Proliferation was significantly inhibited and mononuclear-macrophage differentiated and matured, showing a significant increase in phagocytosis and an increase in non-specific esterase activity. The morphology of cells showed changes consistent with differentiation. Compared to the changes in the neutrophil and lipid profiles of HL-60 cells before and after differentiation induced by GM3, the significant increase in CDH is an important factor in the differentiation of HL-60 cells into mononuclear macrophages induced by GM3. Characteristics, CMH and CQH also increased significantly.
