目的比较正常妊娠和子痫前期、子痫患者胎盘组织血小板源性生长因子(PDGF)-A mRNA、PDGF-AA的表达和血浆PDGF-AA蛋白水平,探讨PDGF-A与子痫前期发病的关系。方法对子痫前期和子痫患者43例、正常妊娠20例,分别用RT-PCR检测胎盘绒毛组织PDGF-A mRNA表达水平,用免疫组化SABC法结合计算机显微图像分析软件,检测胎盘绒毛组织PDGF-AA蛋白的表达水平,用ELISA检测血浆中PDGF-AA蛋白表达水平。结果子痫前期、子痫组及正常妊娠组胎盘组织的PDGF-A mRNA和PDGF-AA蛋白表达,病例组均高于正常组,且与病情轻重呈正相关;病例组血清PDGF-AA蛋白水平均高于正常组,也与病情轻重呈正相关。结论子痫前期和子痫患者胎盘组织及血浆高度表达的PDGF-A可促进动脉粥样硬化形成,可能在子痫前期患者的胎盘血管病变中起重要作用,并参与子痫前期的发生和发展。
Objective To compare the expression of platelet-derived growth factor (PDGF) -A mRNA, PDGF-AA and plasma PDGF-AA in normal pregnancy and preeclampsia and eclampsia, and to explore the relationship between PDGF-A and the pathogenesis of preeclampsia. Methods 43 cases of preeclampsia and eclampsia and 20 cases of normal pregnancy were collected. The expression of PDGF-A mRNA in placental villi was detected by RT-PCR. The expression of PDGF-A mRNA in placental villi was detected by immunohistochemical SABC combined with computerized microscopic image analysis software. PDGF-AA protein expression level of plasma PDGF-AA protein expression by ELISA. Results The expressions of PDGF-A mRNA and PDGF-AA protein in placenta of preeclampsia, eclampsia and normal pregnancy group were higher than those in normal group, and positively correlated with the severity of disease. The serum PDGF-AA protein levels Higher than the normal group, but also with the severity of the disease was positively correlated. Conclusion Placenta and placenta highly expressed PDGF-A in preeclampsia and eclampsia can promote the formation of atherosclerosis, which may play an important role in the pathogenesis of placental vasculopathy in preeclampsia and participate in the occurrence and development of preeclampsia.
