目的建立超高效液相色谱法(UPLC)测定杜仲中主要活性成分松脂醇二葡萄糖苷(pinoresinol diglu-coside,PDG)的方法。方法采用Acquity C18BEH(1.0 mm×100 mm,1.7μm)UPLCTM色谱柱,以乙腈-水为流动相,使用Waters Acquity UPLC system进行测定。结果 PDG的线性范围为4.655~465.5μg,R2=0.999 9,平均回收率为99.4%,RSD=1.7%。结论本方法准确、简便、高效、重现性好,可用于杜仲中PDG的定量测定。同时UPLC在分析强保留、高分离度要求、成分复杂、大批量的样品时具有非常明显的优势,尤其适合对中药复杂体系的分析。
Objective To establish a method for the determination of pinoresinol diglu-coside (PDG), the main active ingredient in Eucommia ulmoides, by ultra performance liquid chromatography (UPLC). Methods The Acquity C18BEH (1.0 mm × 100 mm, 1.7 μm) UPLC (TM) column was used with acetonitrile-water as the mobile phase and determined using a Waters Acquity UPLC system. Results The linear range of PDG was 4.655 ~ 465.5μg, R2 = 0.999 9, the average recovery was 99.4% and RSD was 1.7%. Conclusion The method is accurate, simple, efficient and reproducible. It can be used for the quantitative determination of PDG in Eucommia ulmoides. At the same time, UPLC has obvious advantages in analyzing samples with strong retention, high resolution, complex components and large quantities, and is especially suitable for the analysis of complex Chinese medicine system.
