本文报导了以聚乙二醇为分离剂的血浆胰岛素放射免疫测定法,并对该法主要实验条件和可靠性作了检验。结果显示:聚乙二醇的最佳浓度为15%(w/v),以4℃温育20小时的标准曲线为优,本法的效果与双抗体法相似,其精确性:批内变异CV6.7%,批间变异CV12.4%;准确性:回收率75.5~107.4%;灵敏度:0.03~0.31微单位/0.1毫升血浆。以上结果均符合胰岛素放射免疫分析的要求。
This article reports the plasma insulin radioimmunoassay using polyethylene glycol as a separating agent and tests the main experimental conditions and reliability of the method. The results showed that the optimum concentration of polyethylene glycol was 15% (w / v), and the standard curve incubated at 4 ℃ for 20 hours was excellent. The effect of this method was similar to that of the double antibody method. The accuracy was: intra-assay variation CV6.7%, Inter-assay CV12.4%; Accuracy: Recovery 75.5 ~ 107.4%; Sensitivity: 0.03 ~ 0.31 micro-units / 0.1 ml of plasma. The above results are in line with the requirements of insulin radioimmunoassay.
