目的:建立雀脑药材中丙氨酸含量测定方法,并对不同采集时间的样品进行比较.方法:采用柱前衍生化HPLC法,色谱柱为Elite C18 ( 200 mm×4.6 mm,5 μm),流动相A为乙腈-水(1∶1),流动相B为0.05 mol/L醋酸钠缓冲液(pH=6.4),梯度洗脱,检测波长为360 nm,流速为1.0 mL/min.结果:丙氨酸含量在0.04~0.2 μg范围内线性关系良好,平均加样回收率为99.56%,RSD%为1.67%,不同批次雀脑药材中丙氨酸的含量为3~5 mg/g.结论:丙氨酸的柱前衍生化HPLC含量测定方法重复性好,灵敏度高,可用于雀脑的质量控制.“,”Objective: To establish a method for determination of alanine in sparrow brains by pre-column derivatization and reversed-phase high performance liquid chromatography and compare samples in different collection times. Methods: The chromatographic column was the Elite C18 (200 mm×4.6 mm, 5 μm). The mobile phase A was acetonitrile-water (1∶1) and the mobile phase B was 0.05 mol/L, with sodium acetate buffer (pH= 6.4) and gradient elution. The detection wavelength was 360 nm, and the flow rate was 1.0 mL/min. Results: The linear relation of alanine content determination was good in 0.04-0.2 μg, with average recovery rate of 99.56% and RSD% of 1.67%. The content of alanine in sparrow brains of different batches was 3-5 mg/g. Conclusion: The method for determination of alanine in sparrow brains by pre-column derivatization and reversedphase high performance liquid chromatography shows good repeatability and high sensitivity and it can be used for quality control of sparrow brains.
