目的:优化紫草多糖的提取工艺,探测其分子量组成。方法:采用正交实验方法,以总糖含量为评价指标,对紫草多糖提取工艺进行筛选;将提取得到的紫草多糖进行纯化分离,经体外药效学实验表明其中一组分有抗人乳头瘤病毒(HPV)的作用,用高效凝胶渗透色谱方法(HP-GPC)测定该组分分子量的组成。结果:综合考虑,最佳提取工艺为煎煮3次,加水15倍量,煎煮时间1 h;该多糖的重均分子量分别为28 746,4 877,数均分子量分别为27 336,1 152。结论:优选得到的工艺经济、简单、稳定、可行。
OBJECTIVE: To optimize the extraction process of Lithospermum polysaccharide and to detect its molecular weight composition. Methods: The orthogonal test was used to determine the total sugar content of polysaccharides extracted from Lithospermum. The purified polysaccharides from Lithospermum Phellodendri were separated and purified by in vitro pharmacodynamics test. One of the components was anti-human The composition of the molecular weight of this component was determined by high performance gel permeation chromatography (HP-GPC) using the effect of papillomavirus (HPV). Results: Considering synthetically, the best extracting process was boiling for 3 times, adding 15 times amount of water and boiling time for 1 h. The weight average molecular weights of the polysaccharides were 28 746 and 4877 respectively, and the number average molecular weights were 27 336 and 152 152 . Conclusion: The optimized process economy is simple, stable and feasible.
