目的观察S100B蛋白在孕期不同阶段铅暴露下大鼠胎盘中的表达特征。方法 108只大鼠随机分为4组,于孕期不同时期饮服0.025%醋酸铅溶液;原子吸收光谱法测定孕末期血铅水平;免疫组化法测定胎盘组织中S100B蛋白的表达。计量资料组间比较采用单因素方差分析,计数资料组间比较采用卡方检验。结果实验组血铅水平均高于0.483μmol/L,与对照组比较差异有统计学意义(F=12.10,P=0.000);S100B蛋白主要表达于大鼠胎盘细胞滋养层细胞、蜕膜细胞及血管壁细胞胞质中,实验组阳性表达率高于对照组(52.94%),且孕期全程铅暴露组(93.33%)高于孕早期(87.50%)和孕晚期铅暴露组(80.00%),差异有统计学意义(χ2=18.62,P=0.029)。结论 S100B蛋白的表达与孕期铅暴露大鼠血铅水平相关,推测其在铅致胎盘的损伤中起重要作用。
Objective To observe the expression of S100B protein in rat placenta during lead exposure at different stages of pregnancy. Methods 108 rats were randomly divided into 4 groups, drinking 0.025% lead acetate solution during different periods of pregnancy. Blood lead levels were measured by atomic absorption spectrometry. The expression of S100B in placenta was detected by immunohistochemistry. Measurement data were compared between groups using one-way ANOVA, count data between groups were compared using chi-square test. Results The level of blood lead in the experimental group was higher than 0.483μmol / L, which was significantly different from the control group (F = 12.10, P = 0.000). S100B protein was mainly expressed in the placental trophoblastic cells, decidual cells and In the cytoplasm of blood vessel wall, the positive expression rate of the experimental group was higher than that of the control group (52.94%), and the whole lead exposure group (93.33%) during pregnancy was higher than that in the first trimester (87.50%) and the third trimester lead exposure group (80.00% The difference was statistically significant (χ2 = 18.62, P = 0.029). Conclusion The expression of S100B protein is related to the level of blood lead in lead exposed rats during pregnancy, suggesting that it may play an important role in the lead-induced placental injury.
