本研究采用健康雄性SD大鼠,青年组(3月龄)20只,老年组(26月龄)40只,通过Moris水迷宫行为学测试,将老年大鼠分为老年记忆减退组(简称老年减退组)和老年记忆正常组(简称老年正常组)。用免疫细胞化学、组织化学双标技术定量分析内侧隔核-斜角带(septummedialis-diagonalbandofBroca,SM-DB)中神经生长因子受体(nervegrowthfactorreceptor,NGFR)、一氧化氮合酶(nitricoxidesynthase,NOS)阳性神经元各亚群。老年减退组大鼠和老年正常组大鼠相比较,SM、斜角带垂直支(verticalDB,vDB)、斜角带水平支(horizonalDB,hDB)中NGFR-NOS双标神经元数量明显下降(P<001),其下降幅度为2884%,2333%和2035%;NGFR单标神经元在SM、vDB和hDB中也呈明显下降,其下降值为4043%,4324%,3306%;NOS单标神经元的数量较老年正常组有显著下降(P<005),下降幅度分别为1724%,2000%和1860%。相关分析表明,受试大鼠逃避潜伏期与双标神经元、NGF?
In this study, healthy male Sprague-Dawley rats were used in this study. Twenty young adults (aged 3 months) and 40 senile adults (aged 26 months) were divided into three groups according to Moris water maze behavioral test: Diminished group) and normal memory group (referred to as elderly normal group). Immunocytochemistry and histochemical double labeling were used to quantitatively analyze the expressions of nerve growth factor receptor (NGFR), nitric oxide synthase (NOS) in septum medialis-diagonal band of Broca (SM-DB) Subpopulations of positive neurons. The numbers of NGFR-NOS double-labeled neurons in vertical axis (verticalDB, vDB) and horizonalDB (hDB) in the aged group were significantly lower than those in the aged group <001), which decreased by 2884%, 2333% and 2035%, respectively. NGFR single-labeled neurons also showed a significant decrease in SM, vDB and hDB, with a decrease of 4043% , 4324% and 3306% respectively. The number of NOS single-labeled neurons decreased significantly (P <005) compared with the normal control group (1724%, 2000% and 1860%, respectively) %.
