目的 经体外实验评价紫杉醇对人腺样囊性癌细胞ACC- 2的辐射增敏作用 ,初步阐明其机制。 方法 用克隆形成法检测不同时间 -浓度条件紫杉醇和 /或放射作用后的人腺样囊性癌细胞 ACC- 2细胞存活分数 ,计算辐射增敏率 (SER)。用流式细胞仪分析不同时间 -浓度条件下紫杉醇对细胞周期和细胞凋亡的影响。 结果 紫杉醇对 ACC- 2细胞具有体外增殖抑制作用。 10 ,10 0 ,10 0 0 nmol/ L紫杉醇作用 2 4 h后 SER分别为 1.2 9,1.6 6和 1.2 3。细胞暴露于紫杉醇后发生 G2 / M期阻滞 ,紫杉醇作用时间和浓度越高 ,G2 / M期细胞比率也越高。 10 0 ,10 0 0 nmol/ L 紫杉醇作用 2 4 h后 G2 / M期细胞比率明显升高。10 nmol/ L 紫杉醇作用 2 4 h细胞凋亡率为 2 0 .7%。 结论 一定条件下紫杉醇对人腺样囊性癌细胞 ACC- 2有体外辐射增敏作用。紫杉醇的辐射增敏作用可能是一个多机制事件 ,紫杉醇的 G2 / M期阻滞作用和诱导细胞凋亡作用在其中起重要作用
Objective To evaluate the radiosensitization effect of paclitaxel on human adenoid cystic carcinoma cell line ACC-2 in vitro and to clarify its mechanism. Methods The survival rate of human adenoid cystic carcinoma cell line ACC-2 after paclitaxel and / or radiotherapy was tested by clone formation method. The radiation sensitization rate (SER) was calculated. The effects of paclitaxel on cell cycle and apoptosis under different time and concentration conditions were analyzed by flow cytometry. Results paclitaxel on ACC-2 cells in vitro proliferation inhibition. SER at 10, 10 0 and 10 0 0 nmol / L paclitaxel after 24 h were 1.2 9, 1.6 6 and 1.2 3, respectively. G2 / M phase arrest occurred after exposure to paclitaxel. The higher paclitaxel time and concentration, the higher G2 / M phase cell ratio. After cells were treated with paclitaxel at 10 0 and 10 0 0 nmol / L for 24 h, the percentage of cells in G2 / M phase was significantly increased. The apoptosis rate of cells treated with 10 nmol / L paclitaxel for 24 h was 20.7%. Conclusions Paclitaxel can sensitize human adenoid cystic carcinoma cells ACC-2 to radiation in vitro under certain conditions. Radiosensitization of paclitaxel may be a multi-mechanism event in which paclitaxel’s G2 / M arrest and induction of apoptosis play an important role
