目的:研究参芪扶正注射液联合顺铂对人胰腺癌PANC-1细胞增殖、诱导凋亡的影响,并讨论其作用机制。方法:常规培养细胞,采用参芪扶正注射液和顺铂分别单独及联合作用于PANC-1细胞,于倒置显微镜下观察细胞形态的变化;MTT法检测不同浓度的参芪扶正注射液和顺铂单独及联合作用PANC-1细胞后的抑制率;流式细胞术检测细胞凋亡率,ELISA法检测凋亡因子Caspase-9、Fas和Survivin的含量。结果:参芪扶正注射液和顺铂均能有效抑制PANC-1细胞增殖,并诱导其凋亡,两药联合应用具有协同效应,可促进Caspase-9、Fas的表达,抑制Survivin的表达。结论:参芪扶正注射液联合顺铂能协同抑制PANC-1细胞的增殖,并诱导其凋亡,其作用机制与上调Caspase-9、Fas及下调Survivin因子的表达有关。
Objective: To study the effect of Shenqi Fuzheng injection combined with cisplatin on the proliferation and apoptosis induction of human pancreatic cancer PANC-1 cells and its mechanism. Methods: Conventional cultured cells were treated with Shenqi Fuzheng injection and cisplatin, respectively, separately and in combination on PANC-1 cells. The morphological changes of cells were observed under an inverted microscope. Different concentrations of Shenqi Fuzheng injection and cisplatin The inhibition rates of PANC-1 cells alone and in combination were detected by flow cytometry. The levels of apoptosis factors Caspase-9, Fas and Survivin were detected by ELISA. Results: Both Shenqi Fuzheng injection and cisplatin could effectively inhibit the proliferation and induce the apoptosis of PANC-1 cells. The combination of the two drugs had synergistic effect, which could promote the expression of Caspase-9, Fas and inhibit the expression of Survivin. Conclusion: Shenqi Fuzheng injection combined with cisplatin can inhibit the proliferation and induce the apoptosis of PANC-1 cells in a dose-dependent manner. The mechanism is related to the up-regulation of Caspase-9, Fas and down-regulation of Survivin.
