目的:愈来愈多的证据显示Caspase家族蛋白酶在细胞凋亡执行程序中发挥重要功能。辐射可诱发肿瘤细胞凋亡,分离、鉴定辐射诱发肿瘤细胞凋亡中Caspase家族成员,是肿瘤细胞凋亡调控机制关键所在。方法:根据已知的Caspase家族蛋白保守的氨基酸序列,设计简并引物,逆转录-聚合酶链反应(RT-PCR),分离、鉴定参与辐射诱发细胞凋亡基因。结果:Caspase家族蛋白酶特异的抑制剂能阻抑射线诱发的细胞凋亡,并从照后6hU937细胞分离出caspase-3基因。结论:Caspase参与辐射诱发的细胞凋亡,caspase-3基因是射线诱发细胞凋亡的枢纽元件。
PURPOSE: There is growing evidence that caspase family proteases play an important role in the apoptotic executive program. Radiation can induce tumor cell apoptosis, and to isolate and identify radiation-induced Caspase family members in tumor cell apoptosis, which is the key point of tumor cell apoptosis regulation mechanism. Methods: Degenerate primers and reverse transcription - polymerase chain reaction (RT - PCR) were designed based on the known amino acid sequences of Caspase family proteins. The genes involved in radiation - induced apoptosis were isolated and identified. RESULTS: Caspase-specific protease inhibitors inhibited radiation-induced apoptosis and caspase-3 gene was isolated from U937 cells 6h after irradiation. Conclusion: Caspase is involved in radiation-induced apoptosis. Caspase-3 gene is a pivotal element of radiation-induced apoptosis.
