观察淋巴细胞培养中结构染色体变化是检测辐射与化学诱变剂在体内或体外诱发遗传损伤的一种常规应用方法。一般认为应分析第一次分裂中期(FDM)标本以避免不稳定畸变减少和后继分裂的“衍生变化”。许多人介绍,得到第一次有丝分裂的最适宜时间是48小时,但也有人证明在这个时间可能已有大量的第二次分裂中期(SDM)。本文报告第一次与第二次有丝分裂结构染色体畸变(SCA)频率和类型的比较研究。在1例偶然受很高剂量~(192)Ir照射的8岁小儿研究诱发染色体畸变,并对患Fan-coni贫血的3例同胞(5,6和8岁)观察自发染色体畸变。淋巴细胞培养是应用McCoy培养基,除常规加入胎牛血清与PHA外,另添加BrdU(5μg/ml),培养时间为48小时。为观察SCA,每例分析100
The observation of structural chromosomal changes in lymphocyte culture is a routine method of detecting genetic damage induced by radiation and chemical mutagens in vitro or in vivo. It is generally considered that the first metaphase (FDM) specimens should be analyzed to avoid “derivatization changes” in the reduction of unstable aberrations and subsequent divisions. Many people explained that the optimal time to get the first mitosis is 48 hours, but others have also shown that there may be a large number of second metaphase (SDM) at this time. This article reports the first and second mitotic chromosomal aberration (SCA) frequency and type of comparative study. In an 8-year-old pediatric study with incidental high doses of 192Ir irradiation, chromosomal aberrations were induced and spontaneous chromosomal aberrations were observed in 3 siblings (5, 6 and 8 years of age) with Fan-coni anemia. In lymphocyte culture, McCoy medium was used. In addition to routine addition of fetal bovine serum and PHA, BrdU (5 μg/ml) was added and culture time was 48 hours.
