目的:探讨C-Met和脆性组氨酸三联体蛋白在卵巢肿瘤中的表达及意义。方法:采用免疫组化SP法检测51例卵巢癌、16例卵巢交界性肿瘤、18例卵巢良性肿瘤、10例正常卵巢组织中脆性组氨酸三联体和C-Met蛋白表达。结果:C-Met蛋白在正常卵巢组织中无表达,在卵巢良性肿瘤、卵巢交界性肿瘤及卵巢癌组织中的阳性表达率分别为22.2%,37.5%,66.7%,卵巢癌与正常卵巢组织、卵巢良性肿瘤、卵巢交界性肿瘤比较差异有统计学意义(P<0.05)。脆性组氨酸三联体在卵巢癌组织的阳性率低于正常卵巢组织和卵巢良性肿瘤,差异有统计学意义(P<0.05)。在卵巢癌组织中,随组织学分级的增加和淋巴结的转移,C-Met表达阳性率逐渐上升,脆性组氨酸三联体表达阳性率逐渐下降,差异有统计学意义(P<0.05);C-Met和脆性组氨酸三联体表达呈负相关性(r=-0.382,P=0.006)。结论:C-Met和脆性组氨酸三联体基因突变或缺失在卵巢癌的发生、侵袭和转移中可能起重要作用,C-Met和脆性组氨酸三联体联合检测有助于卵巢肿瘤的早期诊断、早期治疗及生物学行为的评估。
Objective: To investigate the expression and significance of C-Met and fragile histidine triad in ovarian tumors. Methods: Immunohistochemical SP method was used to detect the expression of fragile histidine triad and C-Met protein in 51 cases of ovarian cancer, 16 cases of ovarian borderline tumor, 18 cases of benign ovarian tumor and 10 cases of normal ovarian tissue. Results: The positive expression rates of C-Met protein in normal ovarian tissues, ovarian borderline tumors and ovarian cancers were 22.2%, 37.5%, 66.7% respectively. There was no significant difference in the expression of C-Met protein among normal ovarian tissues, Ovarian benign tumors, borderline ovarian tumors were significantly different (P <0.05). Fragile histidine triad in ovarian cancer tissue was lower than normal ovarian tissue and benign ovarian tumors, the difference was statistically significant (P <0.05). In ovarian cancer, with the increase of histological grade and lymph node metastasis, the positive rate of C-Met expression gradually increased, the expression rate of fragile histidine triad gradually decreased, the difference was statistically significant (P <0.05); C -Met and fragile histidine triad expression was negatively correlated (r = -0.382, P = 0.006). Conclusion: Mutations or deletions of C-Met and Fragile histidine triad genes may play an important role in the occurrence, invasion and metastasis of ovarian cancer. Combined detection of C-Met and Fragile histidine triad contributes to the early stage of ovarian cancer Diagnosis, early treatment and assessment of biological behavior.
