为研究五味子乙素(Sch-B)对二氧化硅致大鼠矽肺中基质金属蛋白酶(MMP-2)和其抑制物基质金属蛋白酶组织抑制因子(TIMP-2)的作用,采用暴露式气管内注入SiO2混悬液法建立大鼠矽肺模型,染毒后1d开始灌胃给予Sch-B干预治疗,药物干预3d、7d、14d和28d后处死取其肺组织.应用HE、Masson染色法观察肺组织病理改变及肺组织胶原纤维变化;通过ELISA法对大鼠肺组织中不同时间点的MMP-2、TIMP-2蛋白的表达情况进行了研究.结果表明:Sch-B对矽肺组织纤维化病变有明显的改善,胶原纤维明显减少.Sch-B对二氧化硅致大鼠矽肺中不同时间点肺组织MMP-2的表达均有一定程度的抑制作用,早期肺泡炎时可一过性提高TIMP-2的表达,起到抑制MMP-2对肺组织损伤的作用,后期抑制TIMP-2的表达,减缓肺组织纤维化的发生.
To investigate the effect of schisandrin B on matrix metalloproteinase (MMP-2) and tissue inhibitor of matrix metalloproteinase-2 (TIMP-2) in silica-induced silicosis in rats, Silicosis model was established by injecting SiO2 suspension, and the rats were treated with Sch-B by intragastric administration on the 1st day after the drug exposure. The lungs were sacrificed on the 3d, 7th, 14th and 28th after the intervention. HE and Masson staining were used to observe the lung Histopathological changes and changes of collagen fibers in lung tissue.Expression of MMP-2 and TIMP-2 protein at different time points in lung tissue of rats was studied by ELISA.The results showed that Sch-B could inhibit the pathological changes of silicosis fibrosis Significantly reduced collagen fibers.Sch-B on silica-induced silicosis at different time points in the expression of MMP-2 in lung tissue have a certain degree of inhibition, early alveolitis can transiently increase TIMP -2, play a role in the inhibition of MMP-2 on the lung tissue damage, late inhibition of TIMP-2 expression, slow down the occurrence of lung fibrosis.
