目的观察番茄红素(Lycopene)对寰枢椎失稳老年小鼠脑的保护作用。方法小鼠寰枢椎周围注射30%乳酸溶液30μl/次,1次/w,连续3w,制作颈椎失稳模型。用番茄红素2个剂量组即5、2.5mg/kg连续1次/d灌胃观察100d,水迷宫测试小鼠记忆能力;酶联免疫分析法(ELISA)测定脑组织TNF-α、IL-1β、IL-6含量;免疫组织化学法(SABC)观察海马CA1区蛋白β-淀粉样蛋白(Aβ)的表达。结果与模型组比较,Lycopene高剂量组小鼠,水迷宫实验错误次数显著减少(P<0.01);脑组织TNF-α、IL-1β、IL-6含量降低显著(P<0.01,P<0.01,P<0.05);脑海马CA1区Aβ表达介于正常对照组和模型组之间。结论Lycopene对寰枢椎长期乳酸堆积所致老年小鼠记忆能力减退具有一定的改善作用,其机制与Lycopene降低脑组织TNF-α、IL-1β、IL-6含量及抑制Aβ蛋白的生成有关。
Objective To observe the protective effects of Lycopene on the brain of aged atlantoaxial instability mice. Methods The cervical instability model was made by injecting 30μl lactic acid solution 30μl / time, 1 time / w around the atlantoaxial axis for 3 weeks. The mice were sacrificed at 2, 5, and 2.5 mg / kg doses of lycopene once a day for 100 days. The memory ability of mice was measured by water maze. The levels of TNF-α and IL- 1β and IL-6 in the hippocampus of rats. The expression of β-amyloid protein (Aβ) in hippocampal CA1 region was detected by immunohistochemistry (SABC). Results Compared with the model group, the number of errors in the water maze test was significantly decreased in the Lycopene high dose group (P <0.01), and the levels of TNF-α, IL-1β and IL-6 in the model group decreased significantly , P <0.05). The expression of Aβ in hippocampal CA1 region was between normal control group and model group. Conclusions Lycopene may play a role in improving memory deficits in aged mice with long-term lactic acid accumulation of atlantoaxial spondylosis. Its mechanism may be related to the decrease of brain TNF-α, IL-1β, IL-6 and the inhibition of Aβ production by Lycopene.
